The pandemic outbreak of coronavirus disease 2019 (COVID-19) across the world has led to millions infection cases and caused a global public health crisis. Current research suggests that SARS-CoV-2 is highly contagious spreads rapidly through communities. To understand mechanisms viral replication, it imperative investigate replicase, huge protein complex comprising up 16 nonstructural associated host proteins, which most promising antiviral target for inhibiting genome replication transcription. Recently, several components replicase in have been solved provide basis design new therapeutics. Here, we report crystal structure nsp7+8 tetramer, comprises two copies each representing nsp7's full-length C-terminus nsp8 owing N-terminus proteolysis during process crystallization. We also identified long helical extension flexible N-terminal domain nsp8, preferred interacting with single-stranded nucleic acids.
